This is a very interesting question because there have some recent changes on who owns the most Hermes Birkins in the world. Rosmah Mansor, the second wife of Najib Razak, former prime minister of Malaysia, is claimed to have the largest Birkin collection in the world. In 2018, Malaysian police found a total of 272 Hermès bags, worth nearly US$13 million, after seizing the family's three apartments in Kuala Lumpur.

However since 2018, socialite Jamie Chua and superstar Victoria Beckham have been getting VERY close to those numbers that Mrs. Mansor use to have in her closet. We have heard from sources that Ms. Chua and Mrs. Beckham have well over 100 Hermes Birkins and I believe they might even have closer to 200 as their collections have added additions over the years.
